My basic suggestion is to work towards a build you have planned for the characters. Choose what you think is the most important skill from that and then go from there. For example, my RDW / GH group:
Ronin:
Idea was to work towards a filled out Overhead tree so Midareba was the priority skill.
DH:
Went with Binds (Gag) and then Climax. In retrospect, I'd probably opt for Viper (less points, similar damage, Force Skill: Dominate superior to Binds) and then Climax.
WM:
My goal was to get Cursecut for Infinite TP ... but that's pointless without heals. Went the route to pick up Cure II 1 first and then backtracked for Cursecut.
Gunner:
Ricochet was the primary goal but, to get SOME Elemental Damage (Conditional Drops, Physical Resist Enemies), I detoured to Voltshot (1 point) on the way to Ricochet.
Hexer:
Prioritized Revenge. Not much else to say here.
As for group composition, it really doesn't matter to be honest. I know I'm probably alone in this thought but EO2 is a pretty easy game as far as the story is concerned. The story has been done with a solo Dark Hunter, I've done the game (start to finish) with a Protector, WM, and DH trio, and I've done the Dragons and Floor 30 boss with a Protector/DH duo ... so pretty much any group can do the story, to be honest.
If you're absolutely dead-set on the "best" team, then:
R DH WM
G H
or
P DH WM
G H
Is probably your best bet. Ronin variant for the main game and possibly Protector one for post-game, though the Ronin one is sufficient. There's topics with builds (such as one) if needed.
