They all respawn once you've killed them with two exceptions:

Floor 23: Respawns after beating the Floor 25 boss.
Floor 25: Does not respawn.

Edit: Forgot to mention, I got Resonance of Fate the other day (on the cheap, impulse buy), game drops you in with no tutorials, leaving it up to you to realize there's some in the pause menu. Not the best course of action for a game with a weird combat system like this. I still haven't figured out...

They do it because the first boss gives more EXP, for less effort (and time), than farming that to max does.

Eh, if it wasn't for the competitive environment, most people playing Pokemon would just do it for the feel-good romp of "starting small and becoming champion" plotline that makes you feel like a winner. Thanks to the fact that anything can become competitive, Pokemon is a lot more popular...
