Ok, I haven't really been trying to plow my way into the endgame activities (assuming they even exist) in the last few days, but I was pretty astounded by what happened on floor 25. So much in fact that when I got a game-over I decided to type down what happened the second time. If anyone gets to this point and feels they didn't follow what happened entirely and want to read it again it'll be in the spoiler below :P

Floor 25 soliloquy(not really) - **MAJOR** storyline spoilers
When you get to the last part.. you'll know because of the huge red door, the voice once again speaks.. and basically the entire story is given to you:

As you struggle to take it all in, the mysterious voice booms from above you.
“Welcome to the inner sanctum of my Heavenly Keep…The King’s Hall.
“We created it in the image of an ancient ark, that we might escape into the skies.
“I fear you will not understand my tale, but listen… Long ago, this world faced destruction.
“The signs were obvious, and so we made plans to flee the perishing earth for the skies.
“But mankind could not adapt to the drastically altered environment below.
“I continued my never-ending research on life to save them. But some were not satisfied.
“They wished to die on their home soil… to return to the false lands on the dead earth.
“I performed experiment after experiment to save those who remained faithful to me…
“But the secrets of life are obscure and arcane. Little time remained to find a breakthrough.
“Thus did I discard my human form, transferring my mind to a body that would last forever.
“In this way, I continued my research to protect my flock from all manner of calamities.
“That research continues today. The Grail of Kings… source of that much-sought immortality…
“It is not yet fully functional. Genetic warping occurs, and men become something entirely other.
“If she had remained with me… Her specialty was genetic research… perhaps…
“No. The past is dead, and it is folly to speculate. I alone will discover man’s salvation!
“I will not let you take the Grail of Kings from here! It is vital to mankind’s future!
“Perish and become dust before my power as the Overlord!”

Fight ensues
There was something pretty peculiar though that reminded me of a certain other RPG: (More spoilers)
After you beat the first Overlord, or his prototype or whatever it is, this happens:

The majestic being known as the Overlord crumbles before you…
As you bask in your hard-won victory, the voice speaks again.
“It seems your power is far greater than I anticipated. But you do not understand…
“My research is still far from complete. Though I have created something of a prototype…
“True eternal life will not be possible until the Holy Grail is finished.
“And so I cannot allow myself to be defeated now.
“But I have a proposition… I will grant you immortality and inhuman strength…
“In return for which, you must leave the Holy Grail with me and depart from this castle.
“Think well… do not all humans desire life eternal and strength unsurpassed?
The Overlord’s offer has been made, and he awaits your answer.
The decision is entirely in your hands…

Return quietly to town? (Yes No)

The first time I played I said no, though was using two retired characters in their 30s, had no items to refill my TP and died honorably :P

Anyways, when I said yes, just to see what would happen, this happened:

Yes: You heed the Overlord’s request and abandon the Holy Grail in favor of returning to town.
Your mission has ended… in a way…
Game Over

 Anyone remember Dragon Warrior/Quest 1, at the very end where you could say yes or no, and the wrong answer would kill you at that point? :P
My War Magus is turning less useful as I go on, but I don't want to give up my healing and I don't want a medic in her place because my War Magus is the second of my two front-liners. *grumble* The War Magus/Hexer combo isn't working as well for me as I wanted it to....

What should I replace the War Magus with?

She's just getting in the way now XP

Ronins are quite powerful. My main party has one in it and he does perform quite well. Midareba is extremely damaging at max. Kinda reminds me of a Survivalist's Apollon, but can be pulled off every turn. Kicker is that it costs 21 TP a use at lv10, but normal physicals do quite well when Midareba isn't needed.

DHs I don't have too much of a clue on their usefulness, but I do use a Medic. She's there as a healing battery/frontline fighter. Makes traversing a floor easier without having to buy tons of Medica IIs every visit to town.

Medics still have Caduceus. Joy. My Medic will have Caduceus in 2 levels (She's at 30).

As for me...I still haven't gone to 13F :P. I read on GameFAQs about retiring and found out that it's a good idea to retire lv30 characters now for the stat boosts and skill points (Stat boosts now apply to every single stat and not just one stat like EOI), then wait until 70 to do it again.

As a result, I retired my main party and releveled them back up to 30. The extra skill points help for sure. The stat boosts are nice as well.

Will be going to 13F for sure tonight unless I find something small and nitpicky enough about my party to rest them for the...oh boy, I lost count how many times I've rested up to this point xD.
